Son of John Spottiswoode (d. 1728) and Helen Arbuthnott (ca. 1675 - 1741). Half-brother (on mother's side) to Walter Macfarlane. Married (1740) to Mary Thomson (daughter of John Thomson of Charleton).

Spottiswoode was at Macfarlane's in London when Boswell visited on March 13, 1763.
